Heteroduplex DNA with nick 5' to mismatch

The daughter DNA strand (upper) contains a mismatched base (red).
The strand discrimination signal, which probably corresponds to strand discontinuities at the replication fork, is located near the mismatch. In vitro, PCNA and RFC proteins are bound to the strand discontinuity that must be present in the DNA substrate.
