Mismatch in heteroduplex DNA with nick 5' to mismatch and nicks around mismatch bound by MutSalpha-MutLalpha complex

[Homo sapiens]

Note that
  • Although in this figure MutSalpha-MutLalpha complex is still located at the mismatch site after introducing the nicks, most likely the MutSalpha-MutLalpha leaves the mismatch site before introducing the nick, probably by sliding movement. This issue, however, is not resolved yet.
  • Physical interactions between PCNA and MutSalpha-MutLalpha complex may take place at this step. Such interactions may help to direct DNA nicking to the daughter strand.
